Description:

Pst I is a type II restriction enzyme that is commonly used in molecular biology research to manipulate DNA fragments. It is derived from the bacterium Providencia stuartii and cleaves DNA at the recognition site 5′-CTGCA/G-3′ to generate DNA fragments with 3′-cohesive termini. Pst I is catalytically active as a dimer and has a molecular weight of 69,500. The enzyme exhibits star activity under certain conditions such as low ionic strength, high enzyme concentration, glycerol concentration >5%, or pH >8.0.

Applications:

  1. Molecular cloning: Pst I is commonly used in molecular cloning techniques to insert genes into plasmids.
  2. Restriction site mapping: Pst I can be used to map restriction sites on DNA molecules.
  3. Genotyping: Pst I can be used for genotyping studies to identify genetic variations.
  4. Southern blotting: Pst I can be used in Southern blotting techniques to detect specific DNA sequences.
  5. Restriction fragment length polymorphism (RFLP): Pst I can be used in RFLP analysis to study genetic variations.
  6. SNP analysis: Pst I can be used to analyze single nucleotide polymorphisms (SNPs).
